Further improvement of phosphite dehydrogenase thermostability by saturation mutagenesis.
Phosphite dehydrogenase represents a new enzymatic system for regenerating reduced nicotinamide cofactors for industrial biocatalysis. We previously engineered a variant of phosphite dehydrogenase with relaxed cofactor specificity and significantly increased activity and stability. Here we performed one round of random mutagenesis followed by comprehensive saturation mutagenesis to further impr...
متن کاملDimerization mutants of alcohol dehydrogenase of maize.
Two ethyl methanesulfonate-induced mutants of the alcohol-dehydrogenase gene in maize have been analyzed. The enzyme subunits produced by the mutants seem to be structurally altered in such a way that the subunit specified by the null allele Adh(1) (0(5657)) does not dimerize with "wild-type" subunits, and that specified by Adh(1) (S(296)) does so only to a limited extent.
متن کاملArabidopsis NAD-dependent isocitrate dehydrogenase mutants
NAD-dependent isocitrate dehydrogenase (IDH) is a TCA cycle enzyme that produces 2oxoglutarate, an organic acid required by the glutamine synthetase/glutamate synthase cycle to assimilate ammonium. Three Arabidopsis IDH mutants have been characterized, corresponding to an insertion into a different IDH gene (At5g03290, idhv; At4g35260, idhi; At2g17130, idhii). Analysis of IDH mRNA and protein s...
متن کاملThermostability of temperature-sensitive folding mutants of the P22 tailspike protein.
Temperature-sensitive folding mutations (tsf) of the thermostable P22 tailspike protein prevent the mutant polypeptide chain from reaching the native state at the higher end of the temperature range of bacterial growth (37-42 degrees C). At lower temperatures the mutant polypeptide chains fold and associate into native proteins. The melting temperatures of the purified native forms of seven dif...
